Normal placental development relies upon the immune tolerance, trophoblast proliferation and invasion, and angiogenesis. Neuropilin-1 is a protein that in humans is encoded by the NRP1 gene. Recent studies have demonstrated that NRP1 plays versatile roles in immunity, angiogenesis, axon guidance, cell survival, migration, and invasion. Our previous studies have shown existence of expression of NRP-1 mRNA in the placenta, and showed dynamic changes: rising in early pregnancy, becomes lowest in late pregnancy, suggesting a potential role of NRP-1 in the pathogenesis of diseases associated with dysregulated placental growth. In this study, we determine the effect of NRP-1 on immunity, trophoblast invasion and angiogenesis in vitro, and in vivo. The information from this study might serve as a basis for future developmental therapeutics of placenta-related diseases.
胎盘是体内唯一不存在神经的特殊器官,有趣的是,神经调节分子往往在胎盘形成发育过程中具有多种生物学效应。免疫耐受、滋养细胞增殖侵袭和胎盘血管生成是胎盘形成发育的三个重要环节。神经纤毛蛋白-1不但调节神经系统的发育,还参与机体免疫调节、肿瘤细胞功能的调节和脉管系统的生成。本项目拟将NRP-1引入胎盘研究领域,我们前期研究显示NRP-1 mRNA在胎盘组织中存在表达,且呈动态变化,早孕期最高,晚孕期最低。推测NRP-1是胎盘形成发育的重要调节因子。本研究拟从体外、在体等层次研究NRP-1在胎盘免疫调节、滋养细胞功能和血管生成三个方面中的作用及机制,为探索胎盘发育寻找新的研究途径和新的理论依据。
胎盘是体内唯一不存在神经的特殊器官,有趣的是,神经调节分子往往在胎盘形成发育过程中具有多种生物学效应。本项目将NRP-1引入胎盘研究领域,我们的研究显示Neuropilin-1在绒毛组织和胎盘组织中存在表达,且呈动态变化,早孕期最高,晚孕期最低。主要表达于滋养细胞和内皮细胞。进一步研究相关病理妊娠患者胎盘组织Neuropilin-1表达情况,发现FGR、PE患者表达明显下降,而前置胎盘和胎盘植入患者表达上升,这些结果提示Neuropilin-1可能参与了胎盘生长发育。细胞学实验进一步证实,下调Neuropilin-1的表达,滋养细胞增殖、侵袭以及管腔形成能力均明显下降,胎盘微血管内皮细胞得出相同的结论。我们的研究证实Neuropilin-1是胎盘形成发育的重要调节因子。这将为探索胎盘发育寻找新的研究途径和新的理论依据。
