中药传统药性、功效与现代药理活性、化学成分之间难以有机结合,是制约中药中西医结合研究的瓶颈问题之一。本项目在前期研究基础上,以伞形科辛味中药川芎、藁本、当归,独活、蛇床子两组为对象,以与活血化瘀相关的药理活性――舒张血管、解痉、抗血小板聚集等为指标,通过辨识有效成分,选取主要活性成分,遵循试验设计-药效试验-数学建模(模型修正)的程序,寻求化学成分组合与多种药理活性之间的关联性,分析成分组合产生血管活性的整合机制,进而深刻揭示"成分组合-活血功效"的关联性。.本项目以伞形科中药且药性基本相同的中药为对象,围绕活血功效,在有限、可控的范围内揭示传统药性与成分、活性的关联规律,不仅对于阐释中药药性的科学内涵,而且对实现传统认识与现代研究之间的有机结合具有重要意义。
中药是药性的载体,化学成分是中药药理活性的物质基础,揭示中药药性-药理效应-化学成分之间的关联规律,对于阐释中药药性的科学内涵,构建基于传统药性的中药质量控制模式和指导中药新药创制,具有重要意义。. 本项目在前期研究基础上,通过建立数学模型,对伞形科辛味中药川芎、藁本、当归的主要活性成分藁本内酯、阿魏酸、丁基苯酞,独活、蛇床子的主要活性成分蛇床子素、欧前胡素、异欧前胡素进行配比优化,并对预测结果进行验证,分别观察两组3种成分组合对离体血管的舒张作用活性,寻求化学成分组合与多种药理活性之间的关联性。在此基础上,探讨成分组合产生血管活性的整合机制,进而深刻揭示“成分组合-活血功效”的关联性。. 研究结果表明,蛇床子素,异欧前胡素,欧前胡素三种成分组合与阿魏酸、丁基苯酞、藁本内酯三种成分组合均对离体血管具有舒张作用,三种成分之间具有协同增效的作用。这种舒张作用靶点在血管内皮和血管平滑肌。血管内皮机制是通过NO-cGMP途径产生,血管平滑肌机制与抑制平滑肌细胞外Ca2+内流相关。.本研究从三个成分、一个模型(离体血管),两个靶点(血管内皮和血管平滑肌)入手,构建了三种成分组合与血管活性的数学模型,确定了最优配比,提出了适合中药配比优化的方法。对深入研究中药药性-药理效应-化学成分之间的关联规律,创新中药质量评价模式、研发中药组分或成分中药具有重要意义。
