More than 30 years have passed since Stevia rebaudiana Bertoni had been introduced successfully as an important sugar plant into China. Now the planters are confronting a severe degeneration of seed strains and big diversity in the content of steviol glycosides of Stevia. .This research proposal is to develop a sensitive and fast immunoassay of stevioside (ST) and rebaudioside A (RA) from stevia leaves through nationwide collection of stevia breeding practice. Hybidoma technique-based monoclonal antibodies of anti-ST and anti-RA will be prepared and used to set up an optimized enzyme-linked sorbent assay (ELISA), in order to support the screening of the large samples of stevia leaves in early elite selection of stevia breeding.
作为重要糖料资源植物的甜菊(Stevia rebaudiana Bertoni)自成功引种我国迄今已逾30年。目前甜菊品种已经出现严重品种退化、主要糖苷类含量(甜菊苷ST和莱鲍迪苷A,RA)差异极大,成为我国利用这一重要植物资源与产业发展所面临的重要课题。因此亟需尽快建立快速、准确的ST和RA测定方法以因应甜菊提高产量、品质改良和早期选优工作的需要。.申请者拟针对甜菊主要糖苷ST和RA制备其相应的单克隆抗体,建立快速灵敏的免疫分析技术,从而筛选出具有高ST和高RA或高RA/ST比值的植株或品系,为提高甜菊育种效率、从量与质两方面改良甜菊品质、开展甜菊育种中早期选优,推动甜菊产业在我国的良性发展提供理论和实验技术支撑。
