The patients with the brain tumor, which received irradiated treatment by using gamma knife, were induced often the brain oedema and harmful consequence. The aim of the present study is that by using neurobiological technique, to research mechanism of the brain oedema induced by gamma knife, and providing metholds by drug. The unilateral caudate-putamen nuclei (CPN) of rat brain were irradiated by different dosage of gamma rays ( from 10 Gy to 100 Gy), and were survived for 3 months, or were irradiated by using 100 Gy and survived from 30 min to 3 months. The brain of experimental rats whether were the brain oedema, and the response of neurons and neuroglias were investiged. The present study discoved that: (1) the brain oedema was not found, (2) clear response of the neurons, astrocytes, microglias and endothelium in irradiated areas and some non-irradiated areas as such the medullary visceral zone were observed. Immunohistochemical reaction to Fos-, GFAP-, HSP70- OX42- and S100 was increased significantly more and appeared time course. Six articles were published ( 2 articles in internatioal journal, 4 articles in China), and obtained good comments from other scientists.
本课题采用免疫组化、免疫电镜、细胞培养以及分子生物学的方法,从超微结构、细胞和分子水平探讨葡萄糖转运体、水转运体、自由基一氧化氮和血脑屏障抗原在r-刀照射引发大鼠脑水肿机制中的作用,同时对水肿变化做长程观察,并初步探讨相关拮抗和药物的抗水肿作用,以了解脑水肿的发生机理,为临床治疗和预防提供理论依据。
