Herba Epimedii is one of the most popular traditional Chinese medicines. Because of the richness in flavonoid constituents and many kinds of pharmacological activity, increasing attention has been paid on Herba Epimedii in recent years. At present, however, the raw materials of Herba Epimedii rely mainly on wild resources, resulting in problems such as the shrinking of wild resources, the confusions of varieties and unstable qualities of Herba Epimedii. In this project, SSRs and ILPs as well as SNP markers discovered by restriction site associated with DNA sequencing (RAD-seq) will be used to construct the high density genetic linkage map of Epimedium based on an F1 population derived from a cross between E. sagittatum (high flavonoid content) and E. leptorrhizum (low flavonoid content). According to the flavonoid contents examined in Epimedium leaves for three consecutive years, QTLs controlling flavonoid contents will be identified. In addition, markers tightly linked with flavonoid contents in Epimedium will also be obtained through developing markers in the confidence interval of major QTL. The project aims to reveal the heredity of flavonoid contents, which will highlight great implications in elucidating the mechanism of its biosynthesis and regulation. Also, the obtained molecular markers closely linked with flavonoid content will provide useful information for molecular breeding of Epimedium cultivars with high flavonoid content, and will effectively promote the sustainable utilization of the wild resources of Chinese Herba Epimedii.
淫羊藿是我国常用中药材,因富含黄酮类成分而具有多种药理活性,备受国内外学者的高度重视。当前淫羊藿药材主要依靠野生资源,存在资源萎缩、品种混乱、质量参差不齐等问题。本项目拟选用已构建的亲本(箭叶淫羊藿和黔岭淫羊藿)间类黄酮含量存在显著差异的淫羊藿F1分离群体为研究对象,利用SSR、RAD-seq、ILP技术检测亲本和分离群体的基因型,构建淫羊藿高密度遗传连锁图谱。通过对亲本及F1分离群体连续三年叶片类黄酮含量的检测,结合淫羊藿高密度遗传图谱,定位淫羊藿药效物质类黄酮含量的QTL位点,开发与类黄酮含量紧密连锁的分子标记。本项目的实施将为解析淫羊藿类黄酮含量变异规律、阐明其合成调控机理奠定基础,对淫羊藿分子育种和选育类黄酮高含量的淫羊藿优良品种具有重要的理论意义与应用价值,将有效促进我国淫羊藿资源的可持续利用。
淫羊藿是我国常用中药材,因富含黄酮类成分而具有多种药理活性,备受国内外学者的高度重视。通过本项目的实施,获得了一批特异性强的EST-SSR引物,其中 4 对引物用于杂交子代的筛选;构建了具有 13,211 个SNP标记,分为6个连锁群,总图距为 11962.9 cM 的整合遗传图谱,其中父本遗传图谱具有 5083 个遗传标记,总图距 11962.99 cM,母本遗传图谱具有 266 个SNP遗传标记,总图距 754.62 cM。与此同时,定位到表型 4 个QTL位点,分布于 2 个连锁群;定位到叶片光谱表型 6 个QTL位点,分布于 4 个连锁群;定位到类黄酮含量相关的 6 个QTL位点,分布于 2 个连锁群;定位到总黄酮含量相关的 3 个QTL位点,分布于 3 个连锁群。本研究对淫羊藿分子育种和培育类黄酮高含量的淫羊藿品种具有重要的理论意义与应用价值,将有效促进我国淫羊藿资源的可持续利用。
