The CDPK (calcium-dependent protein kinase ) gene is a kind of peculiar plant genes. It has an important role in the signal transduction pathways of plant stress. We have cloned a corn CDPK gene and have preliminary analysis of its expresses and shown significantly drought-resistant function in the over-expression of the CDPK gene in the Yeast transformations. On the our previous study basis the project will have conduct the further research of the corn CDPK gene function. First through constructing the different plant transformation carriers of the CDPK gene and transformating into the wild types and mutants of Arabidopsis thaliana and corn at the same time, we will test the transgenic plants phenotype and resistance. Furthermore we will study the corn CDPK gene effects about the over and lack expression of the plant in participating in the plant adversity stress and regulation pathways. Via the yeast two hybrid system and the ZmCPK12 protein phosphorylation we will be sure the adjustment factors of upstream and downstream about the ZmCPK12 gene in participating in the signal transduction pathways of plant stress. As the result of the research will help us understand the role of the CDPK gene in plants growth and the adversity signal transduction, and provide a new evidence and theoretical basis in order to cultivate the new crops about high efficient and stress resistance.
CDPK基因是一类植物特有基因,在植物逆境胁迫信号转导途径中具有重要作用。本项目以已经克隆的一个玉米CDPK基因为基础,以已经对它初步的表达分析和转化酵母表现出抗旱作用的明显提高为依据,对该玉米CDPK基因进行进一步的功能研究。构建该基因不同植物转化载体,转化拟南芥的野生型和突变体,同时转化玉米;检测转基因植株的表型以及抗逆性,研究该玉米CDPK基因是否参与逆境胁迫反应的调控通路;研究该玉米CDPK基因在玉米中过量表达和缺失表达后对玉米胁迫反应调控通路的影响;通过酵母双杂、蛋白磷酸化等实验,确定ZmCPK12在干旱胁迫信号转导通路中的作用及信号转导通路中上下游的调节因子,这一研究将有助于我们了解该基因在植物生长发育和逆境信号转导中的作用,并为我们进一步了解植物对逆境胁迫的复杂的信号转导、基因表达调控体系提供新的证据,以期为培育高效高效、抗逆作物新品种提供新的途径和理论依据。
