In the development of chronic pain, the ectopic discharges from damaged neurons arre main sources of the spontaneous pain and chronic pain. The abnormality of expression patern and function of ion channels have arosen great interest. Hyperpolarization activated channels (Ih) were found widely distributed in the central nervous system, their basic functions are maintaining the rest membrane potential, participating rhythimic acvivities and thereby influence the neuronal excitability. The present grant aims to explore the role of Ih in the mechanisms of chronic pain. Whole cell patch clamp, molecular biological and behavioror methods are conbined together for the invistation. In neuropathic rats with lumbar 5 and 6 (L5 and L6) nerver ligation, it was found that the threshold potential of dorsal root ganglion (DRG) neurons decreased, and the duration of the action potential increased. In addition, Ih mRNA expression in the medium- and small- sized DRG neurons increased when co-cultured with substance P (SP). In conclusion, research results from this grant give primary evidences that Ih may be involved in the abnormal discharges of neurons in the chronic pain condition, and therefore, bocomes a promising therapeutic target for the treatment of chronic pain.
为阐明炎症引起痛觉过敏及痛觉超敏的发病机理,本研究采用膜片钳(电压钳、电流钳、膜电容的测定)记录技术、及结合单细胞PCR的方法,在急性分离慢性炎症痛模型大鼠的DRG神经元,及与SP、CGRP、PGE2长期共培养的DRG细胞上,观察对组织、细胞兴奋性变化节律起决定作用的HyperpolarizationActivatedCationCurrent(Ih)通道在DRG神经元上的变化规律及所起的作用。
