The environment of salt mine belongs to extreme environments. Microorganisms inhabit in extreme environments have very special physiological and metabolic mechanism. Extremphiles could produce active compounds with novel chemical structure. Screened by antibacterial activity screening, 73 active fungi were obtained from 523 fungi isolated from three Yunnan salt mines (Qiaohou salt mine, Yipinglang salt mine and Kunming salt mine). Through the chemical diversity screening, 16 fungi selected from 73 fungi were brought into further screening. The final 3 fungi had not only rich chemical diversity, but also exhibited a variety of anti-fungal and anti-bacterial activities. By studying the proposed project, the bioactive compounds with novel structure could be found from secondary metabolites of 3 fungi of Yunnan salt mines. A new source of lead compounds for drug discovery and essential experimental data can be achieved by this work. The development and application of halophilic fungi in Yunnan salt mines also will be made.
盐矿环境属于极端环境。生存在极端环境中的微生物,其生理与代谢机制极为特殊,产生结构新颖的活性化合物的概率较高。从3个云南盐矿(乔后盐矿、一平浪盐矿和昆明盐矿)分离到的523株真菌,通过抗菌活性筛选得到73株活性菌株;再通过化学多样性筛选,选取16株真菌进入复筛;最终选取3株既具丰富的化学多样性,又具抗多种真菌和细菌活性的耐(嗜)盐真菌。拟通过本项目的研究,从这3株云南盐矿耐(嗜)真菌次生代谢产物中发现结构新颖的活性化合物,为药物发现先导化合物提供新的来源,并提供基础数据,为云南盐矿耐(嗜)盐真菌的开发应用提供理论支撑。
盐矿环境属于极端环境。生存在极端环境中的微生物,其生理与代谢机制极为特殊,产生结构新颖的活性化合物的概率较高。从3个云南盐矿(乔后盐矿、一平浪盐矿和昆)分离到的523株真菌,通过抗菌活性筛选得到73株活性菌株;再通过化学多样性筛选,选取16株真菌进入复筛;最终选取3株既具丰富的化学多样性,又具抗多种真菌和细菌活性的耐(嗜)盐真菌。本项目从3株云南盐矿耐(嗜)盐真菌次生代谢产物中共分离得到73个化合物,最终鉴定了52个化合物的结构,其中新化合物4个,已知化合物48个,结构类型包括麦角甾类、环肽类、内酰胺类、蒽醌类、酚类及其他类型,并对新化合物的抗菌、抗肿瘤活性进行了研究,获得了云南盐矿真菌次生代谢产物的化学多样性信息和基础数据,为云南盐矿耐(嗜)盐真菌的开发应用提供理论支撑。
