The title of this study is: The actions of active components of Huangqi or/and Zhimu on the production of beta-amyloid and on apoptosis. The study has been completed and the proposed aim has been achieved..By [35S]-methionine incorporation experiment and western blot analyis, it was found that AST but not the flavone from Huangqi could markedly inhibit the production of Ab in primary cultured cortical neurones and PC12 cells. ZMS from Zhimu did not show similar effect. Taking the result of Western blot as an example, when the culture media of 48 to 72 hours after addition of drugs were collected and analysed for their Ab relative quantities with the amount of vehicle group as 1, it was found that ZMS group was 1.02±0.15,AST group was 0.64±0.07,while ZMS+AST group was 0.23±0.11. The differences among the three groups were very signifcant. These results are very interesting, since they are in accordance with the Ziyin Yiqi (replenishing vital energy and vital essence) theory of traditional Chinese medicine commonly applcable for treating AD. In view of the importance of Ab in the pathogenesis of Alzhemer's disease, many laboratories are right now searching synthetic drugs for inhibiting Ab production. But the search from tranditional Chinese Medicinal herbs has not yet been reported.In vitro determination of apoptosis-related genes and in vivo Tunnel examination both revealed that either AST (or total saponin from Huangqi) or ZMS could inhibit cell apoptosis. Whether combined use of active components of Zhimu and Huangqi could benifit more has not been revealed clearly. May be more sensitive method should be useful. Both ZMS and total saponin from Huangqi significantly inproved learning and memory in AD model. This is evidently related to the protection effect of these drugs on cortical neurones. However since the importance of apoptosis in Alzhermer's disease is still in argument in literature and the apoptosis of the pathalogical model used in this study was mild, whether the improvment of learning and memory could be completely attributed to the inhibition of cell apoptosis remained to be clarified.
