Bactrocera dorsalis(Hendel) (Diptera:Tephritidae) is an important pest of fruit and vegetables in the world. However, our understanding of component of sex pheromone for courtship and mating in B. dorsalis is still limited. The traditional application of the component identification of B.dorsalis sex pheromone is ineffective. Based on the previous study, a member of Odorant-Binding Proteins (OBP) family, BdorOBP19c is highly and almost exclusively present in female pheromone gland(rectal gland)in puberty, we will combine multiple techniques including transcript sequencing, in situ hybridization, Immunohistochemistry localization, heterogeneous expression, cellular imaging, behavioral analysis, RNA interference (RNAi) to achieve the following three objectives: 1) to identify the function of pheromone carrier protein; 2) to identify the component of sex pheromone; 3) to screen the novelchemicals for insect controlby employing a combination of pheromone carrier protein structure-based drug design and high-throughput screening approaches.Through molecular genetic studies coupled with RNAi to block the sex pheromone delivery, and chemical analysis, a more scientific comprehensive assessment system for sex pheromonecomponent identification would be established. More importantly, it can provide novel methods of pest control, such as attractant with high efficiency, green environmental protection.
桔小实蝇是国际上重要蔬菜水果害虫。桔小实蝇用以求偶交配的化学通信化合物尚未明确,而传统的化学生态分析技术未能对其性信息素进行有效鉴定,本项目在首次证实桔小实蝇气味结合蛋白BdorOBP19c特异表达于性成熟雌虫性腺(直肠腺)的研究基础上,将综合利用转录组测序、原位杂交、免疫组化、异源表达、细胞成像、行为学研究、RNAi技术和化学分析等技术完成3项研究目标:1)明确桔小实蝇性信息素运载蛋白的功能;2)鉴定桔小实蝇性信息素化合物;3)以性信息素运载蛋白为药物设计模板筛选新型活性配体。本项目通过对桔小实蝇性腺中性信息素运载蛋白的功能鉴定,以RNAi阻断性信息化合物转运通路并结合化学组分差异分析,最终建立一种创新性的性信息素组分鉴定研究方法,并为桔小实蝇防控提供一种新的高效、绿色环保的引诱剂进而达到有效防治的目的。
