It has been practically proved that acupuncture therapies can effectively relieve varieties of acute and chronic pains, whereas the underlying mechanisms still need to be further investigated. Studies have shown that neurotransmitters, including GABA, are involved in acupuncture analgesia (AA) at different CNS levels. As an important component of both GABAergic neurotransmission and pain modulating systems, however, the role of GABA transporter-1 (GAT1) in AA remains unclear. The roles of endorphins and opioid receptors in AA have been widely accepted, and our previous data demonstrated that δ-opioid receptor (δOR) can inhibit the activities of GAT1. In this study, we will use a mouse model with neuropathic pain to investigate the influence of acupuncture at certain acupoints on the function of GAT1, and further examine the gene expression level, phosphorylation level, and intracellular distribution of GAT1. In addition, we will test the analgesic effects of acupuncture on GAT1 knockout mice, which may provide further evidences of the involvement of GAT1 in AA. This study will shed light on the function and mechanism of GAT1 in AA, and provide scientific support for the better understanding of AA.
实践证明针刺疗法能够有效缓解各种急、慢性疼痛,然而其机制尚待深入研究。研究表明包括GABA在内的多种神经递质在CNS各级水平参与针刺镇痛过程,作为GABA能神经传递系统以及体内痛觉调制系统的重要组分,GABA转运体-1(GAT1)在针刺镇痛过程中的作用还不清楚。内啡肽及其受体在针刺镇痛中的作用已被广泛接受,我们的前期研究则表明δ-阿片受体(δOR)能够抑制GAT1的生物活性。本研究中我们将在神经病理性疼痛模型小鼠上检测特定穴位的针刺对CNS中GAT1功能的影响,并进一步检测针刺镇痛过程中GAT1的基因表达水平、蛋白质磷酸化水平及其在细胞内的空间分布、以初步揭示GAT1参与针刺镇痛的机制。此外,我们还将检测针刺对GAT1基因剔除小鼠的镇痛效应,用以反证GAT1在针刺镇痛过程中的作用。本研究将初步阐明针刺镇痛过程中GAT1的作用及分子机制,为深入了解针刺镇痛效应提供新的科学依据。
