草酸钙晶体广泛存在于植物组织中,一些新的研究结果颠覆了人们对其生理功能的传统认识,成为植物学研究的新热点。以往草酸钙晶体的观察手段,无法使其和周围的植物组织形成鲜明对比,往往影响研究结果的科学性。草酸钙晶体属于天然离子晶体,而一般植物组织则是由糖类、脂类、蛋白质等构成的没有均匀性和周期性的非晶体。基于草酸钙晶体和周围植物组织非晶体,在晶体独有的光学特性方面理论上会有显著不同,由此推断,以显微镜的入射光为切入点,是实现高清显示植物组织中草酸钙晶体的技术关键。拟对草酸钙晶体和周围植物组织的光学特性差异从四个方面展开研究:①化学本质、②反射光特性、③折射光特性、④光衍射特性,并进一步在普通光学显微镜的基础上,设计出理想的入射光光源,最终解决高清显示植物组织中草酸钙晶体的关键技术问题。研究结果能够提高草酸钙晶体研究的科学性,也能为中药显微鉴定自动化提供良好的图像数据源。
发现了7种实验材料中草酸钙晶体的数量特征和分布规律;证实了草酸钙晶体和周围植物组织的透射光谱和反射光谱存在代表性差异;研究了显微镜入射光与高清显示植物组织中草酸钙晶体的关系,实现了高清显示植物组织中草酸钙晶体的关键技术。
