lncRNA CTC-564N23.3 is a novel long noncoding RNA that highly expressed in differentiated hMSCs-Ad (Human adipose-derived mesenchymal stem cells) identified by lncRNA microarray. Our previous study found that CTC-564N23.3 was highly expressed in adipose tissue of obese children and overexprssion of CTC-564N23.3 promoted hMSCs-Ad differentiation and increased the accumulation of triglyceride. Furthermore, bioinformatic analysis and experiments suggested CTC-564N23.3 promoted hMSCs-Ad differentiation by downregulating ZBED3. This study will firstly verify the relationship between CTC-564N23.3 and obesity. Moreover,we will investigate the function of CTC-564N23.3 in hMSCs-Ad differentiation by overexpression and knock down strategies. Additionally, we will explore the detailed mechanism of CTC-564N23.3 interacted with ZBED3 during the differentiation of hMSCs-Ad. So far there is no report of human lncRNA CTC-564N23.3 in hMSCs-Ad differentiation, thus, this study have certain creativity and will provide a new potential target for the prevention and treatment of obesity.
人CTC-564N23.3是我们前期通过芯片筛选人脂肪干细胞成脂分化前后差异非编码RNA时发现的、差异高表达于成脂分化后人脂肪干细胞的lncRNA。前期还发现,肥胖者脂肪组织CTC-564N23.3表达显著高于正常体重者,过表达CTC-564N23.3可促进人脂肪干细胞成脂分化;进一步生物信息学分析及实验显示,CTC-564N23.3可下调ZBED3表达,进而抑制Wnt信号通路促进人脂肪干细胞成脂分化。本研究拟分析CTC-564N23.3与肥胖的相关性;采用过表达与敲低策略,明确CTC-564N23.3对人脂肪干细胞成脂分化的影响;以ZBED3为机制主线,论证CTC-564N23.3通过调控ZBED3促进人脂肪干细胞成脂分化的具体分子机制。目前尚无CTC-564N23.3的研究报道,本研究将揭示CTC-564N23.3在人脂肪干细胞分化中的作用与机制,为肥胖的防治提供潜在干预靶标。
肥胖(Obesity)在全球已成流行态势,其可致心血管疾病(CVD)、脂肪肝、2型糖尿病、高血压、血脂异常及某些肿瘤的风险显著增高,并已成为全球第5位的死亡原因。近年来,研究人员也已经认识到lncRNA在肥胖、糖尿病、脂肪细胞分化中的重要性,探索lncRNA在人脂肪细胞分化中的作用与机制无疑将会极大促进对肥胖及相关代谢性疾病的理解。本研究发现lncRNA—CTC-564N23.3在心脏、肝脏、骨骼肌表达水平高,肾脏的表达水平很低,且主要位于人脂肪细胞核中。在人脂肪干细胞成脂分化过程中,lncRNA—CTC-564N23.3的表达量明显上调,诱导分化第1和4天表达上调最明显。通过慢病毒技术在人脂肪干细胞过表达lncRNA—CTC-564N23.3,诱导成脂分化,油红染色结果显示过表达lncRNA—CTC-564N23.3组较对照组脂滴数量增加,体积增大;脂肪细胞分化关键基因FABP4表达上调。采用表达谱芯片技术筛选llncRNA—CTC-564N23.3影响人脂肪干细胞成脂分化的可能靶基因;采用KEGG和GO分析分析该lncRNA可能靶基因的集合和功能区域。结果提示KLF5是lncRNA—CTC-564N23.3影响人脂肪干细胞成脂分化的可能靶基因。进一步,沉默lncRNA CTC-564N23.3对人脂肪干细胞成脂分化与对照组相比有抑制分化的作用。人脂肪组织标本(36例肥胖者,12例正常人)检测发现,肥胖者的lncRNA—CTC-564N23.3的表达量明显的高于正常对照组。lncRNA—CTC-564N23.3的表达量与BMI呈现明显正相关。本研究较充分地获得了人lncRNA—CTC-564N23.3在人脂肪干细胞成脂分化中的功能及与肥胖之间的相关性,获得了较多有用的实验依据,为后续研究提供了新线索、新经验、新依据。
