Epilepsy is a kind of nervous system disease which seriously endangers human health. There is a large amount of evidence that the hippocampus and limbic system of epilepsy patients are severely degenerative and lacking. However, the molecular mechanism is not clear. Cell jiao wu is discovered and proved in recent years a new way of programmed cell death, we preliminary study found that people with epilepsy surgery in the hippocampus NLRP1, caspase 1 level increased significantly, and found that LncRNA MEG3 can restrain inflammatory corpuscle NLRP1 regulatory miRNA - 223. Therefore, we hypothesized that LncRNA MEG3 can adsorb mirna-223 to regulate NLRP3/caspase-1 critical signaling pathway, inhibit the focal death of nerve cells, and control epileptic seizure. This research through the bioinformatics prediction, rt-pcr, Western Blot, dual luciferase report and animal experiment, verify the correlation between nerve cells jiao wu and epilepsy, establish LncRNA MEG3, miRNA - 223, NLRP3/caspase-1 express network. To clarify the inhibitory effect of chaihu oyster soup on nerve inflammation and reduce the mechanism of nerve injury, to provide a new potential target for the treatment of epilepsy in TCM.
癫痫是一种严重危害人类健康的神经系统疾病,大量证据表明癫痫患者海马及边缘系统严重神经元变性及缺失,然而分子机制尚不清楚。细胞焦亡是近年来发现并证实的一种新的程序性细胞死亡方式,我们前期研究发现癫痫患者手术切除的海马组织中NLRP1、caspase-1水平明显提高,并且发现lncRNA-MEG3能够调控miRNA-223抑制NLRP1炎症小体。因此我们推测,lncRNA-MEG3能够海绵吸附miRNA-223调控NLRP3/caspase-1关键信号通路,抑制神经细胞焦亡,控制癫痫发作。本研究通过生物信息学预测、RT-PCR、Western Blot、双荧光素酶报告及动物体内实验,验证神经细胞焦亡与癫痫的相关性,建立lncRNA-MEG3、miRNA-223、NLRP3/caspase-1共表达网络。阐明柴胡加龙骨牡蛎汤抑制神经炎症反应,减轻神经损伤分子机制,为中医治疗癫痫提供新的靶点
癫痫是一种严重危害人类健康的神经系统疾病,大量证据表明癫痫患者海马及边缘系统严重神经元变性及缺失,然而分子机制尚不清楚。细胞焦亡是近年来发现并证实的一种新的程序性细胞死亡方式,前期研究发现癫痫患者手术切除的海马组织中NLRP1、caspase-1水平明显提高,因此我们推测神经细胞焦亡、NLRP1炎症小体与癫痫发生密切相关。本研究进一步阐明NLRP3炎症小体在癫痫发病中的作用。研究发现临床癫痫患者血清存在炎症小体激活的现象。实验研究表明NLRP3炎症小体可以激活caspase-1从而诱导IL-1β分泌,通过抑制NLRP3/caspase-1活性减少脑部神经元损伤,延迟癫痫发作时间,减轻癫痫发作程度。柴胡加龙骨牡蛎汤可以抑制NLRP3/Caspase-1神经元焦亡通路中NLRP3、Caspase-1、IL-18、IL-1β、ASC、GSDMD的基因转录,控制痫性发作。通过下调NLRP3/Caspase-1神经元焦亡通路中NLRP3、Caspase-1、IL-18、IL-1β,控制痫性发作,保护神经元,提高神经元细胞活性。柴胡加龙骨牡蛎汤还能够抑制Caspase-1活化,减轻炎症级联反应,减少神经元和星形胶质细胞的炎性死亡。抑制细胞炎症因子IL-1β的成熟,减少星形胶质细胞的活化,降低谷氨酸的释放。通过提高星形胶质细胞胞膜上高亲和力谷氨酸转运体GLT-1/GLAST的表达,增加谷氨酸转运。降低兴奋性氨基酸Glu含量,升高抑制性氨基酸Gly、GAGB含量,恢复兴奋性氨基酸和抑制性氨基酸的平衡,减轻细胞毒性保护神经元治疗癫痫。本项目初步探讨了神经细胞焦亡与癫痫发生之间的关系。明确了炎症小体、NLRP3/Caspase-1、谷氨酸转运体、神经递质之间的调控作用。阐明了柴胡加龙骨牡蛎汤治疗癫痫的调控机制和作用靶点,为中医药防治癫痫提供新思路、新靶点。
