Atrial fibrillation(AF) is the most common clinical arrhythmia, new research suggests that AF is closely related to the abnormal miR - 328 regulation.In our previous study, we found that the expression of LTCC subunit mRNA and protein in the downstream of the miR-328 decreased significantly, so we hope to further analyze the molecular mechanism of miR-328 in the regulation of AF in AF.At the same time, our study indicated that CV-3 is beneficial for AF, whether the efficacy mechanism is realized by miR-328 is worth exploring.Therefore, our project is going to show the expression of cell and animal level verification of miR-328, Ca2+, CaM, Cav1.2,to explore the relationship between miR-328 and myocardial cell proliferation, apoptosis and ultrastructural changes in AF,and to through the interference and overexpression of miR - 328 system of the preparation to clear the key protein identification;In addition, to verify whether CV-3 can improve the occurrence of AF by regulating the expression of miR-328, and provide important clues for the new molecular drug targets of AF therapy;Objective to analyze the effect of CV-3 on the treatment of AF by anti oxidant, anti-inflammatory and anti apoptosis, and provide the objective basis for the clinical treatment of AF with CV-3.
心房颤动(AF) 是临床常见和严重的心律失常,新进研究表明AF与miR-328异常调节密切相关。本课题组前期研究发现房颤时miR-328的下游因子LTCCα1c亚单位mRNA蛋白表达明显下降;同时发现苗药大果木姜子油(CV-3)具有防治房颤作用,其具体药效机制是否通过miR-328实现有待深入研究。基于此,本项目拟在细胞和动物水平验证miR-328 、Ca2+、CaM、Cav1.2的表达变化;探索miR-328与心房颤动时心肌细胞增殖、凋亡和细胞超微结构改变的相关性;同时通过干扰和过表达miR-328体系的制备明确其关键蛋白身份;验证CV-3是否通过调控miR-328的表达改善AF发生,为AF治疗的新分子药物靶标提供重要线索;解析CV-3通过抗氧化、抗炎和抗凋亡的综合治疗心房颤动作用,为苗药CV-3临床应用提供客观依据。
中文摘要(对项目的背景、主要研究内容、重要结果、关键数据及其科学意义等做简单概 述,800字以内):. 心房颤动是目前临床上最常见的一种快速性的心律失常,也是步入老龄化社会的当代中国所需要面对的巨大的社会问题。本课题组前期试验发现含大果木姜子油为主要成分的米槁胶囊对房颤具有治疗作用,且对心肌细胞LTCC蛋白的表达具有改善作用。本课题以大果木姜子油(CV-3)是否对房颤发生过程中miR-328-CaM-CaV1.2信号通路进行调节以达到治疗作用展开研究,为大果木姜子油(CV-3)临床应用提供客观依据。本实验分别从动物实验、细胞实验、过表达miR-328实验,验证miR-328通过miR-328-CaM-CaV1.2信号通路介导房颤的发生,同时予以大果木姜子油(CV-3)干预。使用WB、qRT-PCR等方法检测造模前后各组蛋白、mRNA及各种细胞因子的表达变化情况。动物实验及细胞实验结果表明,造模后,房颤组miR-328的表达增加,较阴性对照组、房颤+CV-3组及房颤+维拉帕米组的表达有差异(P<0.05),同时房颤组的CaV1.2、CaM蛋白表达与阴性对照组、房颤+CV-3组及房颤+维拉帕米组的蛋白表达有差异(P<0.05);过表达miR-328组中CaV1.2蛋白表达减少、CaM蛋白表达增加,与阴性对照组、过表达+CV-3组、过表达+维拉帕米组对比有差异(P<0.05)。同时,实验结果也显示,与房颤组相比,阴性对照组、房颤+CV-3组、房颤+维拉帕米组中IL-1、IL-6表达均增加,差异具有统计学意义(P<0.05)。根据实验结果可知,miR-328与CaM-CaV1.2信号通路参与房颤的发生与维持,而大果木姜子油(CV-3)对于miR-328介导的房颤有治疗作用。大果木姜子作为贵州十大苗药之一,具有温中散寒、理气止痛和防治心悸的作用,以大果木姜子油(CV-3)为主要成分的中成药已运用于临床治疗房颤,此次实验为大果木姜子油(CV-3)治疗房颤提供了实验理论依据,为苗药大果木姜子进一步开发利用提供客观基础。
