本项目以细菌细胞中抽提的质粒DNA为研究对象,研究特殊DNA拓扑结构。应用凝胶电泳、密度梯度超离心、电子显微镜学及扫描遂道显微法研究DNA拓扑结构实体的物理、化学及结构特性。发现a.游离PUC18DNA超螺旋分子具有线圈型结构,为中空球形颗粒,平均外直径43.8nm,包装比21。发现松驰环形DNA分子也存在线圈型结构,其外直径53.1nm。b.质粒DNA分子内存在特殊的拓扑键,可把分子分隔出花瓣样小环。这种拓扑键可能是把细胞染色体分隔成环的结构因子。c.扫描遂道显微法证实,质质粒pUC18DNA可形成面包圈状超凝集结构,外直径16nm。以上结果是DNA结构研究的最新进展,有重要学术价值。
