The abnormal expression of mucin1 (MUC1) well correlates with tumor cell invasion and metastasis. Cancer antigen 153(CA153) is a detectable circulating anti-MUC1 antibody the peripheral blood of patients. It has been widely used in human medicine as a diagnostic and prognostic marker of the breast tumor. Based on previous studies, our study will investigate the biological characteristics of MUC1 in histocytology, genetics and proteomics, as well as analyze the correlation of MUC1 and other related markers in the diagnosis and prognosis of canine mammary tumor. By means of flow cytometry and cell-sorting technique, biological characteristics of various subsets of canine mammary tumor cells separated on their expression of MUC1 will be compared. We will also down-regulate MUC1 expression by RNA interference and investigate its biological function in tumor invasion and metastasis. The purpose of our study is to clarify the relationship between MUC1 expression and pathological features of canine mammary tumor, provide reference for clinical diagnosis and prognosis, and construct a base for future research.
肿瘤的转移和侵袭行为与肿瘤细胞膜上异常表达的粘蛋白1(MUC1)有关。癌抗原CA153是一种可以在患者外周血循环中检测到的MUC1蛋白,其作为乳腺肿瘤诊断、预后的标记已被广泛应用于人医临床。本课题拟在前期研究的基础上,从组织细胞学、基因和蛋白表达等方面考察MUC1的生物学特点,并分析其与相关标记物在犬乳腺肿瘤诊断和预后方面的相关性。同时,根据MUC1表达情况,借助流式细胞分析和分选技术对犬乳腺肿瘤细胞进行分群,比较不同亚群细胞间的生物学特性,并通过基因干扰沉默MUC1的表达,分析该蛋白参与调控犬乳腺肿瘤侵袭和转移过程中的生物学作用。本研究有望通过对特定标记物定性、定量分析,明确MUC1的表达与犬乳腺肿瘤病理特点的相关性,为肿瘤的诊断、预后评估提供参考,为后续开展靶向治疗研究奠定基础。
研究背景:MUC1基因在乳腺肿瘤的侵袭、转移及病理分级中发挥重要作用,其编码产物癌抗原CA15-3是一种可以在患者外周血循环中检测到的蛋白。这种蛋白作为乳腺肿瘤诊断、预后的标记已被广泛应用于人医临床,但在兽医临床少有报道。.研究内容:.1. 通过ELISA检测乳腺肿瘤患犬血清中CA15-3的水平,应用IHC检测CA15-3在犬乳腺肿瘤组中的表达,并分析CA15-3与肿瘤大小、动物年龄、肿瘤组织分级和预后的相关性。.2. 通过荧光定量PCR检测犬乳腺肿瘤组织中MUC1基因的表达情况,IHC检测MUC1在犬乳腺肿瘤组中的表达情况,分析MUC1与肿瘤大小、动物年龄、肿瘤组织分级和预后的相关性。.3. 构建犬乳腺肿瘤细胞系,筛选表达MUC1的细胞株。.4. 构建MUC1基因沉默细胞模型,分析MUC1基因对犬乳腺肿瘤侵袭和转移能力的影响。.研究结果:.1. CA15-3在乳腺肿瘤患犬血清中含量为13.49±13.04 U/mL,明显高于健康犬(6.07±5.16 U/mL),在不同预后病例中差异显著,CA15-3在犬乳腺肿瘤组织中高表达。.2. 犬乳腺肿瘤组织中MUC1基因高表达,且在不同的恶性分级中差异显著。.3. 成功构建犬乳腺肿瘤细胞系CMT-7364,筛选出表达MUC1蛋白的细胞株。.4. MUC1基因沉默细胞模型的构建正在进行中。.科学意义: CA15-3和MUC1犬乳腺肿瘤诊断和恶性程度评估提供参考,为犬乳腺肿瘤的临床治疗提供理论数据。
